제목 Effect of Alginate and Demineralized Bone Particle Sponges with Chondrocytes for the Articular Cartilage Tissue Engineering
초록 Alginate and demineralized bone particle (DBP) sponges were fabricated by 10, 20, 40 and 80 wt% alginate/DBP hybrid sponges for seeding cartilage cell. Cell proliferation activity was measured via MTT assay. Morphological observation, histology, biological assay for collagen type I, II and aggrecan, and PCR were performed at each time point 1, 2 and 3 weeks. The cell viability was better in 20 and 40 wt% alginate/DBP sponges than other wt% alginate/DBP sponges. Scanning electro microscope (SEM) results showed that more attach cell in the 20 and 40 wt% alginate/DBP sponges with the lapse of time. Dimethylmethylene Blue (DMMB) results showed that the high content of Glycosaminoglycan (GAG) and collagen levels at 20 and 40 wt% alginate/DBP sponges. This study suggested that alginate/DBP sponge may serve as a potential cell delivery vehicle and a structural basis for tissue engineered articular cartilage.
