Polysaccharides which are from various kinds of natural materials are useful for tissue engineering scaffolds due to their biocompatibility, biodegradability and non-toxicity. In this study, we successfully fabricated a modified pullulan hydrogel using enzymatic cross-linking method which is composed of a modified polysaccharide and horseradish peroxidase. The modified hydrogel was analyzed by ATR-FTIR, 13C NMR and 1H NMR in order to confirm the introduction of tyramine groups and coupling with phenyl groups. Rheological properties were also evaluated by adjusting the amount of H2O2 and HRP, concentration of polymer solution and substitution ratio of tyramine group. We investigated human adult fibroblasts’ behavior in DMEM with hydrogel and characterized by MTT assay. |
