Growth Factor-Loaded F-68/PVA/Chitosan Composite Nanoparticles for Cartilage Regeneration |

F-68/PVA/chitosan composite nanoparticles were designed and characterized for the growth factor delivery. TGF-β2, transforming growth factor-β2, was used as a model growth factor. F-68/chitosan composite prepared heparin immobilized F-68 and chitosan molecules via ionic interaction. After that, chitosan modified F-68 solution and PVA solution were mixed with vigorous stirring to form to F-68/PVA/chitosan nanoparticles. For growth factor loading, TGF-β2 dissolved in PBS is mainly adsorbed on the chitosan-modified F-68. To prepare the nanoparticles in the powdery state, F-68/PVA/chitosan composite nanoparticles were freeze-dried. Field emission scanning electron microscopy (FE-SEM), zeta-potential, and particle size analyzer were used to characterize the nanoparticles. The release pattern of loaded growth factor was measured by using ELISA kit, respectively. As a result, with the immobilization of heparin into the F-68/PVA composite, the sustained release of TGF-β2 was accomplished. |
