Sustained Release of VEGF-loaded Nanoparticles with Lecithin Core for Ischemic Disease Therapy. |
초록 |
VEGF loaded nanoparticles were prepared and characterized as a protein delivery system. The lipid core is composed of soybean lecithin and polymeric shell is composed of Pluronic F-68. The freeze-drying of VEGF-loaded lecithin core was performed in the F-68 aqueous solution containing trehalose used as a cryoprotectant to form stabilized core/shell nanoparticles. FE-SEM was used to observe the morphology of core/shell nanoparticles and electrophoretic light scattering was used to observe the particle size and zeta potential. The activity of protein is confirmed by inject into corneal limbus of New Zealand White rabbit (3-3.5kg, male) and myocardial infarction-induced rat model. VEGF was loaded into the core/shell nanoparticles by self-assembly and electrostatic interaction. As a result of release pattern, the core/shell nanoparticles show the sustained release of loaded protein. The core/shell nanoparticles in this study can be utilized as a protein delivery system for angiogenesis. |
