Label-free tomographic imaging is applicable to therapeutic evaluation of targeted nanodrugs in lipid-laden foam cells |
초록 |
Lipid droplet accumulation is a key feature of foam cells and an attractive target for therapeutic intervention in atherosclerosis. However, the availability of non-invasive and quantitative methods is highly limited for use in living foam cells. Here, using optical diffraction tomography, we performed quantitative morphological and biophysical analysis of living foam cells in a label-free manner. We also identified lipid droplets in foam cells by verifying the specific refractive index. Furthermore, through time-lapse monitoring, we precisely and quantitatively evaluated the therapeutic effects of a nanodrug designed to affect the targeted delivery to foam cells. |
