Preparation and Transfection efficiency of Bioreducible gene carrier with chitosan for effective gene delivery |
| 초록 |
To improve gene transfection, release in cytosol was important factor. These for reason, bioreducible gene carrier was prepared by radical polymerization with chitosan and bis-acrylamide cystamine. Then, Its structural analysis was accomplished by 1H-NMR and FT-IR. In addition, geletardation assay showed that pDNA was completely condensed by bioreducible gene carrier. Moreover, It was protected from DNase and released by heparin. Also, transfection assay indicated that polyplex with bioreducible carrier was enhanced more than that without disulfide bond. It means that release of pDNA from polyplex was improved by dissociation of disulfide-bond from bioreducible carrier. into cytosol with glutathion. These results suggest that bioreducible carrier can apply as carrier for gene deilivery.
