제목 Therapeutic applications of photofunctional dendrimer nano-devices
초록 A novel polymeric micelle system, which is formed via an electrostatic interaction of anionic dendrimer photosensitizers (DPs) and poly(ethylene glycol)-poly(L-lysine) block copolymers (PEG-b-PLL), was prepared for the utilization of effective photodynamic therapy. The photodynamic efficacy of the DP was drastically improved by the inclusion into the polymeric micelles, exhibiting a more than two orders of magnitude increase in the photocytotoxicity compared with the free DP, due to the accumulated singlet oxygen production in the intracellular compartment as well as to the modulated intracellular localization related to the micellar structure. On the other hand, a novel ternary complex, composed of a core containing the packaged DNA and an envelope of DP as a light-responsive gene carrier, was prepared. The ternary complex showed more than 100-fold photochemical enhancement of the transgene expression in vitrowith reduced photocytotoxicity.
