A caspase-3 protease biosensor was successfully developed as a model system for the diagnosis of various kinds of diseases related on the initiation and propagation of apoptosis. Capase-3 cleavable site of DEVD connected to a partial sequence of E. coli RNAI was labeled with TAMRA as an optical probe. After binding to the GO by π-π stacking, the quenched fluorescence of 5'-DNA-3'-DEVD-TAMRA was observed to be recovered via the enzymatic cleavage by caspase-3 in apoptotic cells. Furthermore, fluorescence of 5'-DNA-3'-DEVD-TAMRA was also observed in hypoxic ischemia brain and NDMA induced cirrhotic mice. From the results, we could confirm the feasibility of the optical biosensor of 5'-DNA-3'-DEVD-TAMRA/GO complex for the detection of caspase-3 which is the indicator of cell apoptosis. Moreover, the multiple stacking of 5'-DNA-3'-cleavable sites-dyes on the GO will be presented to develop multi-purpose diagnostic sensors for the simultaneous detection of various kinds of biomarkers. |
