제목 Biosynthesis of cadaverine from galactose by reconstruction of metabolic pathways in Escherichia coli
초록 As an alternative way to replace polyamides from the petroleum-based process, biological fermentation of cadaverine has become one of solutions. Although cadaverine has to be fermented with high yield and productivity from diverse sugars rich in biomass for an ideal production, however, most microorganisms are not feasible to metabolize other biomass-derived sugars as efficiently as glucose. Because of it, low growth rate and carbon flux for the production has become the problem. Thus, we engineered Escherichia coli to produce cadaverine from galactose, a promising future feedstock. For doing this, genes related in the production of cadaverine were overexpressed through synthetic expression cassettes. In addition, we inactivated the feedback inhibition and degradation/re-uptake pathways. Finally, the resultant strain produced 8.80 g/L cadaverine with high yield (0.170 g/g) and productivity (0.293 g/L/h). These values were similar to or better than the previous glucose fermentation.
