제목 Preparation of 5-fluorouracil loaded PLGA wafer and release behavior in vitro

Poly(D,L-lactide-co -glycolide) (PLGA)-based wafers offer many advantages, such as the control the drug release rate, good biocompatibility, complete biodegradation. 5-fluorouracil (5-FU) is a water-soluble drug and an antimetabolite of the pyrimidine analog type that is widely used against a variety of solid tumors with usually incorporating into microencapsulation because of its short half-life and its poor oral absorption. The aim of this study was preparation of PLGA wafers containing 5-FU and evaluation of the release kinetics from the prepared wafer in vitro. 5-FU was mixed with PLGA by Rotary evaporation method and the wafer was fabricated by direct compress method. The drug loading amount of the wafer was 10%, 20% and 30% and the size of the wafer was 3mm(diameter) by 1mm(thickness). In vitro release of 5-FU from the wafer was assayed by high performance liquid chromatography (HPLC). Morphological changes and degradation behavior of the wafer was determined by scanning electron microscopy (SEM), mass loss and water uptake observation. The prepared 5-FU loaded PLGA particles were characterized by X-ray diffractometer (XRD) and differential scanning calorimetry (DSC).
