4-cyano-4´-pentylbiphenyl (5CB) in a transmission electron microscopy (TEM) grid was functionalized with a homo PAA-b-LCP (TEMPAA), and a mixed PAA-b-LCP and QP4VP-b-LCP brushes (TEMmixed), and glucose oxidase was immobilized to the PAA chains via coupling agent in the first one while through electrostatic interaction to QP4VP in the later one. The glucose was detected from a homeotropic-to-planar (H-P) orientation change of the 5CB observed through a polarized optical microscope. The utility of both biosensors were compared for glucose detection in term of enzyme stability and reactivity, sensitivity, reusability, and linear dynamic range. The TEMPAA has high sensitivity (0.02 mM) while the TEMmixed has large linear dynamic range (0.5-11 mM). These TEM grid glucose biosensors provides a new way of detecting glucose, with many practical advantages such as the absence of interference from other analytes, the ease of detection, and the selective glucose detection in a complex mixture. |
