제목 Optimization of In Vitro Transcribed (IVT) mRNA Synthesis for Improved Translation Efficiency.​​​
초록 The prospect of IVT mRNA therapeutics can go beyond the limits of plasmid DNA for target protein treatments. It is receiving a lot of attention in the field of gene therapy and regenerative medicine.​ The IVT mRNA template consists of a T7 promoter, a 5′UTR, an ORF, and a 3′UTR. Cap analogues and m​odified rNTP are mainly used toincrease the translation efficiency of IVT mRNA and modulating the immune response. In addition​, removal of dsRNA fragment generated during the IVT process is recommended through various purification methods. We designed GFP and Luc reporter gene constructs for IVT mRNA synthesis and optimized the IVT condition. In our study, modified mRNA prepared with pseudo-UTP and AG-clean cap showed enhanced GFP and Luc expression as compared to the native mRNA. It was confirmed that the synthesis yield and translation efficiency were significantly different depending on the cap analogue.
