In vitro and in vivo artificial cartilage formation usingsurface-modified porous biodegradable polymer scaffolds |

Recently, there are many research approaches in tissue engineering field. Biodegradable aliphatic polyesters such as poly(lactide), poly(glycolide), and their copolymers are very widely used as porous scaffold system for transplanted cell support in regeneration of artificial tissue-engineered organs. Particularly, biodegradable polymer scaffolds with an interconnected open pore structure have been actively studied on tissue-engineered artificial cartilage in clinical applications including plastic and reconstruction surgery. Therefore, optimal bio- and cell compatibility of polymeric material surface play an important role in tissue engineering applications. In this study, we investigated in vitro chondrocyte culture on porous biodegradable polymer scaffolds prepared by using a gas forming technique with nontoxic effervescent mixture and modified with plasma glow discharge treatment for acrylic acid (AA) grafting. Also, we have regenerated neocartilage using transplanted cells in vivo under precultured and non-precultured conditions. The chondrocyte-seeded surface-modified scaffolds were implanted in subcutaneous tissue of nude mouse for predetermined times. And then DNA content and Collagen Type Ⅱ expression from the 3-D cultured scaffolds were evaluated.
