Calcium phosphate (CaP) has been used for the delivery of nucleic acid drugs into cells. DNA or RNA co-precipitates with calcium phosphate at high calcium concentrations, and the precipitates are internalized by cells to release their internal cargos at low calcium concentrations. Also, the solubility of CaP is highly dependent upon pH, the CaP can be useful for pH-responsive release of drugs. However, the irregular size of CaP should be controlled for further utilization of CaP in biomedical applications. In presence of block copolymers with neutral hydrophilic block and negatively charged block, CaP nanoparticles with unimodal size distribution can be obtained. The CaP nanoparticles showed pH- and calcium concentration- dependent stability, which can be very attractive for the controlled release system. We formed PEG-PMP coated CaP nanoparticles containing bisphosphonate drugs for the new therapeutics in osteoporosis. |
