제목 | The Preparation of Polymeric Gene Carrier to Target Artery Wall Cells |
초록 | Non-viral gene carriers, such as Poly-L-lysine (PLL), Polyethylenimine (PEI), can overcome immunogenicity produced by viral vectors in vitro, but their in vivo applications are greatly hampered due to their poor biocompatibility and rapid degradation. On the other hand, chitosan is the second most plentiful biomass and is already known as a biocompatible, biodegradable, and low-toxic material. Chitosan is also considered to be a good candidate for the gene delivery system, since cationically charged chitosan can be complexed with negatively charged plasmid DNA. In this study, Artery wall binding peptide (AWBP) was conjugated to PEGylation chitosan to target arterial wall cells and AWBP-PEG-Chitosan could be used to understand the genetic treatment of cardiovascular diseases. AWBP-PEG-Chitosan was synthesized by the conjugation reaction between the vinylsulfone group of PEG-g-Chitosan and the thiol group of cysteine in AWBP. Characters of the synthesized AWBP-PEG-Chitosan were analyzed through FT-IR, 1H-NMR, zeta-potentiometer, atomic force microscopy (AFM), transmittance electron microscopy (TEM). And characters of the physical and biochemical properties AWBP-PEG-Chitosan/pDNA complexes will be also investigated. |
