제목 Fabrication of protein array on the micropatterned substrate using dendrimer porphyrin and layer-by-layer (LbL) method
초록 A dendrimer-coated surface is one of the promising candidates for the design of effective microarray, because it provides a 3-D architecture with numerous functional groups to immobilize proteins, and can minimize protein denature due to flexible nature of branched arm. Here, to prepare protein microarray with high sensitivity and stability, a dendrimer porphyrin (DP) with 32 carboxyl groups on the periphery was used as a potential building block for immobilizing proteins. By combining photolithography, LbL self assembly and spin self-assembly technique, micropatterned (PEI/PSS)40PEI/DPs multilayer structure with size of 50 um was successfully constructed. Feasibility of immobilizing protein onto DP was confirmed by visualizing localization and patterning of FITC-conjugated casein. Finally, immunoassay was performed using antibody, and it was found that 0.1 20 ug/ml of target antigen could be successfully detected with enhanced fluorescence signal and sensitivity.
