초록 | The optimal pH control profile was calculated with the experimentally developed model and minimum principle of Pontryagin. The calculated pH was abruptly changed from the growth optimal pH to the curdlan production optimal one after ammonium source depletion. Similar to the other polysaccharide productions, two phases existed for the cell growth and product formation. During the cell growth period, the required oxygen amount is high and carbon dioxide production is also high. However, the oxygen requirement and carbon dioxide production rapidly decreased during the curdlan formation period. From this information, the cell states could be determined indirectly. In this research, three kinds of feedback optimal control have been applied for the maximal production of curdlan. Each feedback control algorithm control the pH profiles to the optimal one. Curdlan is an insoluble polysaccharide composed exclusively of -1,3-linked glucose residues, and synthesized by Agrobacterium species and Alcaligenes faecalis under the nitrogen-limiting conditions.Curdlan was also can be used for an admixture to concrete to enhance the fluidity of concrete and has been commercialized by Takeda Chemical Industries Ltd., Japan. There are other polysaccharides used as admixtures of concrete and their competitive price must be minimized to have economic feasibility. Therefore, it is important to increase curdlan productivity by manipulating the environmental conditions and its production cost should be minimized to compete with other polysaccharides.The increase of curdlan production rate decreases the production cost and enhances the profits. Therefore, the optimization of the curdlan fermentation process for maximal production has been of much interest to industry. There are many factors involved in optimizing curdlan production. Temperature, pH, nutrients, agitation, and aeration are the most typical factors. The optimization of agitation speed on the curdlan production has been done by Lee et al. Since cell growth and product formation rates are greatly depend on pH variation, the optimization of pH profiles is also important in maximizing curdlan productivity. |
