In stem cell based therapy, several drawbacks such as low cell survival and engraftment after transplantation exist. Adipose derived stem cells (ADSCs) have been investigated for ischemic diseases. We synthesized pH-sensitive gold-iron nanoparticles (AuFeNPs) to enhance angiogenic paracrine factor secretion of ADSCs. Previously, the concentration of iron (Fe) ion is related to cellular hypoxia and secretion of angiogenic factors such as vascular endothelial growth factor (VEGF) and fibroblast growth factor (FGF). Fe ion delivered to ADSCs using our nanoparticles enhanced angiogenic factor secretion. Fe ion was released from AuFeNPs in the low pH condition of endosomes after endocytosis due to the different reduction potentials between Au and Fe. AuFeNPs treated hADSCs showed enhanced angiogenic factors secretion, compared to hADSCs treated with AuNPs. The novel nanoparticle, as a carrier for pH-responsive intracellular metal ion delivery, may be innovative method for clinical therapy. |
