Catechol-immobilized nanofiber for enhancing attachment and migration of cells |

Catechol has a strong adhesiveness with surface of substrates. Also, the catechol could bind with thiol groups of various amino acids by Michael-type addition at basic condition because the dihydroxy group transform into diketone group. Almost dihydroxyphenyl group has a limitation of adhesiveness after polymerization of catechol. Therefore, in this study, nanofiber was surface-immobilized [CNF] with a single molecule of catechol to enhance cell adhesiveness of the nanofiber. NIH 3T3 fibroblasts were cultured on CNF to confirm a cells adhesion in DMEM without serum at pH 7.4 for 3 h. Among of the nanofibers, CNF showed about 5-fold higher of cells adhesion compared to NF. It was expected that catechol on the nanofiber could induce an adhesion of cells because the catechol interact with thiol-group of surface-proteins on cell membrane. This CNF showed a superior incorporation of thiol groups, also, significantly stimulated adhesion and migration of cells compared with NF. |
