Morbidity and mortality due to influenza could be reduced by development of effective vaccination. Skin immunization is attractive, because the skin is replete with antigen-presenting cells. We prepared arrays of microneedles coated with influenza vaccines including trehalose in the coating formulation which significantly increased vaccine stability during coating. A single dose of influenza vaccine from coated microneedles provided superior protection against lethal viral challenge at the main site of viral replication in the lung, as evidenced by virus clearance below the detection limit. Apart from immunologic advantages, microneedles also offer potential logistic opportunities such as fater, simpler, painless, and suitable for self administration. In summary, our results suggest that influenza vaccine delivery to the skin using microneedle patches may provide a new modality to increase patient coverage and improve immunogenicity of influenza and other vaccines. |
