제목 Large-scale Production and Therapeutic Compound Regulation of Extracellular Vesicles via 3D Stem Cell Bioprocess Using Biopolymers
초록 As a lipid-shielded and nano-sized vesicle retaining medicinal potency equivalent to that of mesenchymal stem cells (MSCs), MSC-derived extracellular vesicle (EV) is preferred over the current MSC infusion therapy with improved safety profiles, considered an ideal therapeutic candidate for regenerative medicine. However, conventional MSC culture methods are problematic to obtain sufficient amounts of EVs for clinical application. In addition, as primed for a targeted disease, desired recruitment of the multifaceted therapeutic compounds in EVs should be addressed. In this talk, I will introduce efficient 3D stem cell bioprocessing techniques using biopolymers, which are 1) a microwell culture platform composed of polyethylene glycol hydrogel that successfully reached significant enrichment of MSC-derived EVs, and 2) 3D-organization of different physical interactions between MSCs and culture matrices using gelatin methacryloyl hydrogel to regulate cytokine inclusions packaging into EVs.
