제목 Preparation of hydrogel microparticles entrapping enzyme immobilized silica nanoparticles.
초록 Silica based nanoparticles were widely used as a support for enzyme immobilization due to high loading capacities and easy surface modification. However, enzymes are easily denatured when they are immobilized on dry substrates such as silica. One solution to maintain enzyme activity for a long time is to use hydrogel matrix with high water content. Although hydrogel is capable of entrapping enzyme without impairing enzyme structure and activity, mass transport limitation and enzyme leaching have limited the use of hydrogel for enzyme immobilization. We prepared PEG hydrogel entrapping enzyme pre-immobilized on the silica nanopartcle. Glucose oxidase was chosen and enzyme activity was investigated by varying the molecular weight of PEG and silica content. Entrapping silica immobilized enzyme provided better mechanical strength of PEG hydrogel and prevented enzyme from leaching out from hydrogel matrix, which allow us to use higher MW of PEG for the improved enzyme-catalyed reaction.
