A novel pH-sensitive fluorescent probe with controllable photothermal effect (IR825-loaded carbonized zwitterionic polymer, FNP-I) as smart drug vehicles for cancer treatment was developed in this study. The "off" fluorescence of FNP-I was activated at tumors (acidic pH) where FNP lost its hydrophobicity and release almost 100% of IR825. The FNP-I are excellent biocompatibility for both normal and cancer cells in vitro. The in vivo tests suggested after IV injection into MDAMB-231-cell bearing mice, the FNP-I was accumulated mostly in the tumors. After exposing to 808 nm laser at 2 W cm-2 for 5 min, the tumor was effectively necrotic compared to control. In addition, the combination of the biosensor and PTT in a single FNP-I led to more effective cancer cell killing than either monotherapy. In conclusion, the smart pH-responsive FNP-I showed high potential for simultaneous cancer diagnosis and treatment. This work was supported by Korea National University of Transportation in 2018. |
