Superparamagnetic iron oxide nanoparticles (SPION) currently have been a surge of interests as their potentials have been demonstrated in biomedical applications such as magnetic resonance imaging (MRI), drug delivery, and therapy (hyperthermia). For clinical MRI applications, it is essential to engineer or passivate the labile surface of SPION with favorable functionalities such as: i) anti-biofouling property to render it resistant to protein adsorption and ii) good dispersibility in physiological medium (i.e., high salt and protein concentrations) without formation of any aggregates during reasonably long time period. Here we report 1) synthesis and characterization of the PEG-Silane polymer-coated SPION and 2) its efficacy as a cancer imaging agent in vivo by using conventional clinical 1.5 Tesla MRI. We have synthesized two kinds of the polymer-coated SPION by two different preparation methods and compared in vitro and in vivo properties of each SPION as a MRI contrast agent. |
