Herein, we prepared melanin@perfluorohexane@5-fluorouracil(MPF)-liposomes for improved treatment outcomes by inducing thermal ablation and drug release due to bubble generation based on the light-to-heat conversion properties of melanin. MPF-liposomes were prepared with a size of 209.61±4.28 nm. Upon laser irradiation, MPF-liposomes exhibited effective temperature rise in vitro and in vivo. In this regard, temperature rise caused a phase transition of PFH to induce dramatic bubble generation, resulting effective drug release behavior. When MPF-liposomes were injected into tail vein of CT26-bearing mouse followed by exposure to NIR laser, temperature of the tumor region was increased to 52℃ and ultrasound contrast significantly increased due to bubble generation. More importantly, tumor growth was effectively inhibited in the MPF-liposomes with laser irradiation group. These results suggest that MPF-liposomes can be utilized as therapeutic agents and drug release-inducing agents. |
