Integration of cell-derived ECM and polymer toward therapeutic applications |

Extracellular matrix (ECM) is a complex but highly organized network of diverse macromolecular components that play a critical role in maintaining tissue homeostasis. To harness the great benefits of ECM toward therapeutic purposes, we have utilized human lung fibroblast-derived ECM (hFDM) that is obtained after decellularization of in vitro-cultured cells. These hFDMs are further processed to make a physical integration with biocompatiable synthetic polymers. This presentation is composed of two separate topics; 1) A novel platform of cardiomyocytes culture and co-culture via fibroblast-derived matrix-coupled electrospun nanofiber, and 2) Fabrication of stretchable ECM cardiac patch and its application to myocardial infarct. Taken together, cell-derived ECM-coupled polymeric biomaterial can provide not only an excellent microenvironment for cell adhesion and differentiation but may reinforce a therapeutic efficacy of human mesenchymal stem cells. |
