Co-delivery of Plasmid DNA and Adjuvant using Degradable Microneedles for Enhanced Cancer Vaccination |

Microneedles (MNs) is considered as a non-invasive manner to deliver the therapeutic agents through stratum corneum painlessly. Among them, dissolving MNs is usually fabricated by biocompatible materials that can dissolve in the interstitial fluid of the skin. In this study, we developed an amphiphilic copolymer Pluronic F127 (F127) based nano micelle to generate the MNs loading the hydrophobic adjuvant R848 and DA3/pOVA polyplex. DA3/pOVA polyplexes and F127/R848 micelles were released in the epidermis/dermis and a portion of F127/R848 micelles were also drained to the lymph nodes, which generates relatively superior cellular and humoral responses. R848 treated MNs group enhanced the OVA specific antibody level and intensity of IFN γ+CD 8 +T cells, which exhibited a remarkable effect on OVA specific cancer vaccine. Therefore, dissolving MNs encapsulating DA3/pOVA polyplex and R848 could be convenient and safe vaccination method with enhanced vaccine efficacy for cancer immunotherapy. |
