제목 Cell encapsulation using thermosensitive chitosan copolymer
초록 Thermosensitive chitosan has been prepared by grafting N-isopropylacrylamide (NIPAAm) to chitosan using ceric ammonium nitrate (CAN) as the initiator. The reaction chemistry and properties of the graft polymers were investigated. Cell compatibility of copolymers have been investigated by cell culture method. Cell-entrapped the chitosan-g-PNIAAm beads or matrix were utilized for cell culture study using fibroblast and mesenchymal stem cells (hMSCs). Cell viability of fibroblast and hMSCs in the chitosan-g-PNIPAAm copolymer gel and alginate gel was investigated. In vitro chondrogenic differentiation of hMSCs within the chitosan-g-PNIPAAm copolymer was investigated. The cell viabilities in both conditions were maintained over 70%. In addition, differentiation into chondrocyte has been shown. Novel chitosan-g-PNIPAAm copolymers have been developed and these copolymers might be very useful as injectables for cell and drug delivery in biomedical and pharmaceutical applications.
