The widely studied strategy of synergetic delivery of siRNA and anticancer drugs for cancer treatment has clinical potentials, but it still has important issues such as delivery efficiency or biocompatibility of the siRNA delivery system. In this study, we present a simple and biocompatible colloidal formulation, i.e. hydrophobically associated multiple monocomplex (HMPlex) (<10 nm in hydrodynamic size) consisting of Bcl-2 gene targeting siRNA, anticancer agent (BZT, benzethonium chloride) and Pluronic F-68 with low charge ratio (N/P = 8). It was demonstrated by in vitro studies using various cell lines that HMPlex containing siRNA and BZT efficiently led to both mRNA down-regulation by siRNA and cell apoptosis by BZT, compared to non-targeting HMPlex with scrambled siRNA and a non-toxic cationic surfactant. |
