Skin, a multi-layered structure such as epidermis and dermis, is mainly constituted with keratinocytes and fibroblasts. It is important that two different cells were cultured in separating environment for preparing an artificial skin. In this study, in order to separately culture the two different cells, hydrogel (22%, w/w) composed of diacrylated Pluronic F127 and glycidyl methaacrylated chitosan (20%, w/w) was prepared in an aim to encapsulate fibroblasts. Also, the hydrogel contained a mixture of basic fibroblasts growth factor (100ng) and heparin (218ng). For separation of two types of cell, the hydrogel was covered surface-immobilized nanofiber with epidermal growth factor because the several nanometers of nanofiber pores can perform a barrier. Then, keratinocytes were seeded on the nanofiber. After cultivation for 28 days, proliferations of the cells were measured by MTT assay. Also, histology and RT-PCR were performed to confirm location and phenotype of the cells. |
