The role of multi segmented metallic nanorod in immune cell culture |
초록 |
Multi segmented nanorods applied to gene and drug delivery due to their unique morphological features. Various ligands can be precisely decorated on the each segment of the nanorods without disrupting adjacent segments by different conjugation chemistry. We electrodeposited the nanorods composed of gold and nickel segments and surface-decorated the nanorods with two different ligands which are recognized by immune cells. Mannose and GRGDS peptide were simultaneously conjugated on the surface of gold and nickel segments, respectively, which can be served as anchorages for antigen presenting cells (APCs) and immature T cells. Conjugation of the ligands was characterized by confocal microscopy analysis. Surface-decorated nanorods with Jurkat cells, immortalized human T lymphocytes, CD11c+ bone marrow dendritic cells from C57B/6 mice were added to the incubation medium. The levels of cytokines such as interleukin (IL)-2 were determined by an enzyme-linked immunosorbent assay (ELISA). |
