In cancer gene therapy, developing a safe and accelerated gene transfer system to deliver nucleic acids have been focussed a lot nowadays. Previous studies have shown polysorbitol modified PEI have improved the transfection efficiency in cancer cells. In cancer cells, microRNA 145 which is a novel tumor suppressor gene involved in cell suppression, invasion and migration are found downregulated. We investigated the polysorbitol-mediated transporter (PSMT) efficiency in the cellular delivery of miRNA 145. Physicochemical characterization and transgene expression capability of PSMT/reporter plasmid DNA nanoparticles were determined. GFP-tagged miRNA 145 delivery with PSMT was confirmed by confocal microscopy and Western blotting. Finally, the expression of a miRNA 145 target protein, c-myc, was determined by Western blotting. Thus our study has shown that microRNA 145 delivered by PSMT in breast cancer cells have reduced cell proliferation much better compared to the control carrier. |
