제목 Bio-reducible nanocarrier for therapeutic microRNA delivery to the treatment of breast cancer
초록 Polysorbitol-based osmotically active transporter (PSOAT) was shown to possess dramatically enhanced transfection activity with reduced cytotoxicity. miR-145 expression levels are usually down-regulated in breast cancer tissues than in their normal counterparts. PSOAT was utilized to deliver microRNA 145 for the treatment of breast cancer. The miR-145 and GFP dual expression reporter plasmid was delivered with bio-reducible PSOAT gene carrier to study the transfection efficiency. The effects of the expressed miR-145 on proliferation, migration and invasion in vitro were analyzed. The repression of the targets of the miR-145 and the downstream signaling pathway was studied with PCR and western blot analysis. The tumor xenograft model was established, and the PSOAT complexed miR-145 was intra-tumorally injected. The tumor suppression after the delivery of particulate microRNA-145 was studied and confirmed with immunohistochemical analysis.
