Electrospun nanofibrous mesh including catechol-modified poly (ethylene glycol) and thiolated poly (lactic-co-glycolic acid) for anti-fouling tissue adhesive |
초록 |
Tissue adhesives using electrospun nanofibrous mesh (NF) can be applied to the suture site after surgery as an ‘anti-adhesive barrier’. In order to achieve anti-fouling effect and adhesiveness to tissue surface, catechol moiety was conjugated to 8-arm poly (ethylene glycol) (PEG). Electrospun PLGA-SH/cPEG NF was prepared by electrospinning with catechol-modified 8-arm PEG (cPEG) (Mw 20,000) and thiolated poly (lactic-co-glycolic acid) (PLGA-SH) (Mw 20,000; lactide:glycolide=75:25) dissolved at concentration of 40% (w/v) in chloroform and methanol mixture (3:1). For cross-linking, sodium periodate (3mM) was treated to NFs and completely dried for overnight. We confirmed cross-linking between catechol moieties and thiol groups by differential scanning calorimety and raman spectroscopy. Cross-linked NF showed good anti-fouling effect on biomolecules and also showed delayed mass erosion rate, which suggests that mechanical strength was increased by cross-linking. |
