siRNA has been received a great attention as a gene therapeutics. However, development of efficient siRNA delivery system is difficult due to low charge density and rigid structure of siRNA. To overcome these drawbacks, we developed a siRNA concatamer system using rolling circle transcription, followed by nanocomplex formation with thiolated glycol chitosan (tGC). The formation of siRNA concatamer and nanocomplex with tGC was investigated by electrophoresis. We further analyzed the physical properties of siRNA concatamer/tGC nanocomplex, such as size, surface charge, and morphology. Our siRNA concatamer/tGC nanocomplex can be a promising candidate for cancer therapy. |
