Preparation of Pegylated-Chlorambucil Prodrug Nanoparticle for Solubilization of Hydrophibic Drug and Enhanced Anticancer Effect |
초록 |
Leukemia has been treated by Chlorambucil (CLB). According to recent studies, CLB reported that has anticancer effect against various cancer cells. However, CLB is limited because of the MDR effect, high toxicity, and low solubility in water. So, we designed pegylated-CLB (PCLB, mPCLB) prodrug by using coupling agents. Their chemical structure was analyzed by 1H-NMR and UV-vis spectrophotometer. In addition, dispersity of them against water with various concentration was confirmed by UV-vis spectrophotometer. Its particle size and morphological property were confirmed by using DLS and TEM. Cytotoxicity and anticancer effect were investigated by using MTT assay. Besides, to confirm their cellular uptake at cancer cell lines, FITC was labeled to prodrug and observed by fluorescence microscopy. These results suggest that they can apply as anticancer agent.
