Preparation and Characterization of Doxorubicin-loaded Redox-sensitive Polymeric Nanoparticle Based on Water Soluble Chitosan for Cancer Therapy |
초록 |
Recently, number of researcher developed to drug carrier in order to delivery of hydrophilic and hydrophobic substance in order to drug delivery in cancer cell. However, To solve these problem, redox-sensitive polymeric micelle was synthesized by coupling reaction between lipoic acid and water soluble chitosan (WSC). Redox-sensitive polymeric micelle has an advantage of trigger-drug release by glutathione (GSH) into cancer cell. Chemical structure of LA-grafted water soluble chitosan (LWSC) was confirmed by 1H-NMR and FT-IR. Its critical aggregation concentration (CAC) was analyzed by fluorophotometer. Also, Doxorubicin (DOX)-loaded WSC (DLWSC) nanoparticles were produced by dialysis method. Moreover, Cytotoxicity and anticancer activity were conducted by using MTT assay. These results suggest that redox-sensitive DLWSC is a superb anticancer agent for cancer therapy. (NRF-2016H1D5A1910499). |
