A new pH-sensitive, biodegradable and injectable hydrogel has been developed for drug and protein delivery which based on poly(ethylene glycol)-poly(sulfamethazine-carbonate-urethane) [PEG-PSMCU]x copolymer. This hydrogel exhibited a sol-to-gel transition in response to change of pH. In sol state at high pH (e.g. pH 8.8), the therapeutics can be easily mixed with aqueous copolymer solution, followed by inject into the body and form an in situ hydrogel depot for sustained release of loaded therapeutics. The hydrogel was characterized using 1H NMR, FT-IR, scanning electron microscopic (SEM), rheological analysis, MMT assay for cytotoxicity evaluation. The results have indicated that the synthesized hydrogel had an interconnected and homogeneously porous structure, cytocompatibility and biodegradability, forming a good complex with positively charged protein. Hence, the novel hydrogel is considered as promising candidate for drug and protein delivery. |
