With the aim of studying the molecular level behavior between Lipopolysaccharide (LPS) and immune protein (LBP, CD14), we used Atomic Force Microscopy (AFM). For regeneration of real in vivo system, LBP and LPS(Salmonella minnesota, Ra,Rc,Re) which has different saccharide region length were immobilized to the AFM tip over chemical spacer linker. On the other hand, lipid bilayer model biomembrane including CD14 was prepared as hydrophobic SAM and lipid layer on gold surface. With these tip and substrate, we measured the binding force between LBP and CD14, LBP-LPS complex and CD14, LBP-LPS complex and CD14 while PMB injection. From our experiment, we inferred the mechanism that LBP-LPS complex meets CD14 in existence of PMB. |
