3D co-culture system in multi-functional hydrogel based scaffolds for enhancing osteogenesis |

Cell encapsulation within hydrogel scaffolds has been studied for tissue engineering application. This study about paracrine effect between mesenchymal stem cells (MSCs) and osteoblasts was systemically investigated in the biopolymer-based three-dimensional scaffolds. For this, osteoblasts were encapsulated in alginate microbeads using electrostatic droplet extrusion. They were co-encapsulated with MSCs in biopolymer bulk hydrogel scaffolds. We have hypothesized osteoblasts can enhance osteogenesis in MSCs by the paracrine effect. Therefore, we have prepared three experimental groups, osteoblasts with MSCs, bone morphogenetic protein 2 (BMP-2) with MSCs and only MSCs in 3D hydrogel culture to compare the efficiency of paracrine effect. Our system developed in this study can be used to study the paracrine effect from the factors released from the MSCs by separating MSCs in bulk hydrogel from alginate encapsulated osteoblasts. |
