An ideal gene carrier requires both safety and transfection effciency. Polyethylenimine(PEI) is a well-known cationic polymer which has high transfection efficiency owing to its buffering capacity. But it has been reported that the PEI has cytotoxicity in many cell lines and the toxicity increases with increasing molecular weight. To reduce toxicity of the PEI, many groups reported "PEGylation" to PEI but the transfection efficiency showed tendency to decrease with an increase of degree of substitutions. In this study, we synthesized a novel degradable copolymer composed of PEI and PEG through reaction of short linear PEI to PEG diacrylate as a cross-linker with degradable ester linkage in physiological condition. The obtained copolymer was characterized by H1-NMR spectroscopy. The copolymer/DNA complex were characterized by measuring particle size, zeta potential and TEM. And we confirmed low cytotoxicity and high transfection efficiency of the complex by performing in vitro cytotoxicity test and transfection experiment. |
