제목 Efficient Gene Delivery by Urocanic Acid-Modified Chitosan

Chitosan is considered to be a good candidate for gene delivery system, since it is already known as a biocompatible, biodegradable, and low toxic material with high cationic potential. However, chitosan has several disadvantages that are poor solubility in water or organic solvents, which can make chitosan difficult to be chemically modified and low transfection efficiency. One of the primary causes of the poor gene delivery is inefficient release of the DNA/polymer complex from endocytic vesicles into the cytoplasm. In this study, to enhance the transfection efficiency of chitosan, water-soluble chitosan (WSC) was coupled with urocanic acid (UA) bearing imidazole ring which can play the crucial role in endosomal rupture through proton sponge mechanism. Its DNA complexation, protection ability and cytotoxicity were characterized, and the parameters that affect DNA transfection efficiency were investigated. It showed great ability to form complexes with DNA and showed better physicochemical properties compared with chitosan. These polymers also had low cytotoxicity and exhibited much enhanced gene transfer efficiency than chitosan itself. Therefore, urocaninc acid-modified chitosan has much potential for safe and efficient gene carriers.
