제목 In situ Forming Hyaluronic Acid Hydrogels for the Treatment of Rheumatoid Arthritis
초록 In situ forming tyramine modified hyaluronic acid (HA-Tyr) hydrogels were successfully developed for the treatment of rheumatoid arthritis (RA) with reactive oxygen species facilitating the hydrogel formation. HA-Tyr hydrogels were prepared for the encapsulation of dexamethasone (DMT) as a model RA drug by radical crosslinking reaction using hydrogen peroxide and horse radish peroxidase. According to in vitro release tests, DMT was sustained released from the HA-Tyr hydrogels up to a month. Intra-articular injection of the in situ forming HA-Tyr hydrogels encapsulating DMT resulted in successful treatment of RA with statistically significant reduction of IL-6, PGE2 and other cytokine levels in collagen-induced arthritis animal models. Histological analysis with hematoxylin & eosin staining also confirmed the therapeutic effect of injectable HA-Tyr hydrogel systems. The recovered cartilage after treatment with HA-Tyr hydrogels containing DMT was comparable to that of normal SD rats.
