Because DNA library screening is generally tedious procedure, studies on simplified screening methods have been actively investigated. In this study, DNA separation was performed by electrophoresis in modified-polyacrylamide-gel, and in-gel expression was performed in electrophoresis gel for direct DNA screening. Electrophoresis gel was modified by Nitrilotriacetic acid (NTA)-moieties which are widely used for capturing histidine-tagged protein by addition of Ni2+ ion. The NTA-functionalized monomer was synthesized by epoxy-amine reaction between glycidyl methacrylate and Nα, Nα-Bis(carboxymethyl)-L-lysine(BCML). |
