Cancer immunotherapeutic strategy via organic-inorganic nanohybrids for priming tumor-associated macrophages toward anti-tumor phenotype |

Directing pro-tumorogenic tumor-associated macrophages (TAMs) near the tumor regions to anti-tumorogenic macrophages has been emerged as an effective strategy to treat cancers. Herein, we developed a cancer immunotherapeutic strategy to re-educate TAM toward anti-tumor phenotype and increase nitric oxide (NO) production for the inhibition of cancer cell propagation. Polyamino acid/MnO2 nanocomplex was designed for the immunotherapeutic agent, which included TAM phenotype-converting cationic polymer and biodegradable MnO2 carrier. Cytotoxicity caused by cationic properties was reduced by surface modification with biocompatible carbohydrates. Also, the manganese ions released from the nanocomplex could not only promote iNOS expression in the macrophages but also provide MR imaging ability. Our results showed notable therapeutic effects of cancer cells, derived from interrelations between macrophages and the components of the nanocomplex. |
