Ascites-driven Protein Corona Enveloped Nanoparticles: Improving Peritoneal Retention, Tumor Penetration and Activatable Drug Release Properties in Peritoneal Carcinomatosis Treatment |

Survival rate and poor prognosis rate among peritoneal carcinomatosis (PC) patients prompted us to fabricate a smart drug delivery system with high anti tumor potential. Herein, we designed paclitaxel (PTX) loaded glutathione (GSH) responsive micelles that can enhance peritoneal retention time by forming a large size aggregates in ascitic fluid, penetrate tumor nodes and could release the PTX specifically in GSH rich environment of cancer cells. Nanoparticles administered in intraperitoneal space enhanced peritoneal retention and low systemic circulation. Also, in vivo studies showed significant inhibition of PC. Immunohistochemically analysed tumor tissue demonstrated the high damage and apoptosis. To conclude, the design of drug loaded nanoparticles which could increase the peritoneal retention by protein corona formation and tumor penetration proved to be an efficient strategy for treating PC. |
