Preparation and characterization of CdSe/ZnS quantum dot encapsulated poly(ethylene glycol)-b-poly(D,L-lactide) micelle nanoparticles |

The final goal of this study is to develop multi-functional organic/inorganic hybrid nanoparticles, applied for both biomedical imaging probes and drug delivery carriers. As an initial step toward this goal, we used a solid dispersion method for encapsulating CdSe/ZnS quantum dots(QD) into poly(ethylene glycol)-b-poly(D,L-lactide) micelles. The size and morphology of the particles were analyzed by using DLS and AFM. The fluorescence intensity of the particles was determined by fluorescence analysis device. The QD-encapsulated particles had 149±11nm of mean size and 121±12 (RFU) of fluorescence intensity. We also tested the cytotoxicity and cellular uptake of the particles. The cell viability was 60~80% in various concentration ranges of particles. We verified cellular uptake of the particles using a FACS analysis and a confocal microcope. Our results suggest that the QD-encapsulated PEG-PLA nanoparticles can be a promising multi-functional nanocarrier for imaging and drug delivery. |
