Regeneration of bone fractures requires new technologies in tissue engineering. Hydroxyapatite(HA) is a synthetic mineral which has similar crystal structure to inorganic compounds of natural bone. In this study, we blended poly(lactic acid-co-glycolic acid) with HA to generate composite nanofibers via electrospinning. Characterization of the nanofibers was performed with SEM, AFM. Our results demonstrated that HA was homogeneously dispersed in the nanofibers. Incorporation of HA on nanofibers didn’t affect cell viability, as examined by MTT assay. The effects of PLGA/HA composite nanofibers on osteogenic differentiation of hMSCs were determined using ALP assay, Calcium assay, RT-PCR, Von kossa staining. Our results showed that the addition of HA on the PLGA nanofibers enhanced calcification of hMSCs. These results indicate that the electrospun PLGA/HA composite nanofiberous scaffolds can be used as a promising substrate for bone regeneration. |
