Non-viral delivery of minicircle DNA to induce myogenic differentiation of human chondrocytes |
초록 |
One of methods for deriving specific therapeutic cellsis lineage conversion from one somatic cell type to another. Also recent study have been focused on non-viral vector delivery with high expression efficiency. Here, We developed a direct conversion methodology for human chondrocytes using electroporation and poly (beta-amino esters) (PBAE) transfection. We delivered minicicle DNA - based reprogramming factor by electroporation followed by PBAE transfection into human chondrocytes. The various transfection results such as facs analysis and fluorescence microscopy are provided and myogenic differentiation was demonstrated with specific gene marker via western blot. |
