Calcium Phosphate Mineralized Self-Assembled Nanoparticles for Enhanced Photodynamic Therapeutic Efficacy |

Calcium phosphate (CaP) mineralization of self-assembled nanoprticles can produce biocompatible, robust nanocarriers that can induce facilitated drug release in intracellular endo/lysosomes. In this study, we describe the potential of mineralized nanoparticles for photodynamic therapy of tumors. The core-shell-corona polymer micelles (PM) of poly(ethylene glycol)poly(L-aspartic acid)poly(L-phenylalanine) (PEG-PAsp-PPhe) was utilized as a nanotemplate for CaP mineralization. In particular, PAsp middle shells with anionic carboxylates serve as a template for CaP mineralization. The CaP mineral layer of CaP-mineralized PM (PM-CaP) can protect efficiently the release of a photosensitizer (PS) at extracelluar environments (pH 7.4) and dissolve in endosomal compartments (pH 5.0) to facilitate the intracellular release of the PS. In vitro cytotoxicities for breast cancer MCF-7 cells were estimated. Intracellular localization was also confirmed using confocal laser scanning microscopy. |
